How To Select Antibodies?

Researchers have taken advantage of the high specificity of antibodies for antigens to gain access to various information.

1) Detection of target molecules

• Antibodies are used to determine the expression level and localization of the desired molecule.

• Applications: Western blotting, immunoprecipitation, ELISA, flow cytometry, immunohistochemistry, etc.

There are many companies available that can provide recombinant rabbit monoclonal antibodies products and services

2) Isolation and purification of target molecules

• Only target molecules are isolated from complex mixtures such as extracts from cells and tissues, which contain large amounts of proteins, nucleic acids, amino acids, hormones, and more.

• Example: Epitope tagged protein purified with antibody-coated agarose

3) Isolation and purification of cells

• Cells expressing cell surface proteins were isolated from the cell mixture using antibodies against the target protein.

4) Functional analysis of target molecules in vivo

• The function of the target molecule is analyzed in vivo by administering inhibitory (or stimulatory) antibodies to cultured cells and experimental animals.

• Example: The antitumor effect of antibodies against the Treg marker GITR (DTA-1) was examined in tumor-bearing mice.

Issues to consider when selecting antibodies

Review the information below to select the appropriate antibody for your experimental design.

1) Name of the antigen

xx in anti-xx is the name of the antigen. First, search using the term "anti-xx (name of the desired molecule)". Also, search using alternative names and abbreviations.

2) Clonality

mAb: monoclonal antibody

pAb: Polyclonal antibody

3) Application

It is important to know whether the antibody is suitable for the intended use. The antibodies in this example can be used for flow cytometry, immunocytochemistry, immunoprecipitation, and western blotting.

4) Conjugation

For secondary antibodies, select the appropriate label for the planned experimental system (fluorescent dyes, enzymes, magnetic beads, etc.).

• Antibody labeling information

 5) Host Species

When selecting secondary antibodies, information about the animal being immunized is important.

• Information on secondary antibodies

• Information about immunized animals
